Effect of Mannitol and Betamethasone on Postischemic Brain Edema in Rats


Abstract
This study was performed to evaluate the effect of mannitol and betamethasone of the postischemic brain edema by recirculation after 30 minutes, 1, 2, 3, and 6 hours, following transient clamping of both common carotid artery for 3 minute and 5 minute of durations.
12 rats were untreated controlled and 24 rats were treated with the combination of mannitol(2 gm / kg) and betamethasone (1 mg / kg) intravenously.
The contents of water, amount of Na and K in the postischemic brain were measured and compared with each group between the 3 minute clamping group and 5 minute clamping group, and between the untreated group and treated group with mannitol and betwamethasone.
The results were as follows
1. The water content of the treated group was significantly decreased in both 3 minute and 5 minute clamping group, comparing with that of the controlled group.
The water content of the 5 minute clamping group was significantly increased than that of 3 minute clamping group.
2. The amount of Na and K were significantly increased in the treated group.
But the difference between the 3 minute clamping group and 5 minute clamping group is insignificant.
3. The amount of Na, K and Na with K in relation to water content of the treated group were higher than that of the controlled group.
From the results, it is suggested that the mannitol and betamethasone might be effective for the prevention and treatment of brain edema.
